Abstract:
Cell therapy is a promising strategy for the treatment of human

diseases. Tracking
the movement and location of living cells both *in vitro* and *in vivo* is
important in various branches of life sciences. Particularly in stem cell
research, robust and reliable tracking of transplanted cells is essential
towards detailed understanding of their development, fate, and contribution
to regenerating tissues. While the first use of cells for therapeutic
purposes can be traced to the 19th century, there has been a lack of
general and reliable methods to study the biodistribution and associated
pharmacokinetics of transplanted cells in various animal models for
preclinical evaluation.

Here, we present a new platform using fluorescent nanodiamonds (FNDs) as
biocompatible and excellently photostable fluorophore for cell labelling
and precise quantification *in vivo*. FND possesses negatively charged
nitrogen-vacancy centers (NV–) which emits far-red fluorescence emission
(~700 nm) with long fluorescence lifetime (~20ns). Its fluorescence is
distinctly different from those of tissue auto-fluorescence enabling
background-free imaging of FNDs-labelled cells in tissue sections by
time-gated technique. Especially, FND is unique of its super-inertness and
magneto-optical property allowing quantitative tracking and quantification
of transplanted cells in animal models. We have demonstrated the technique
with mesenchymal stem cells (MSCs) both in mice and miniature pig.
Moreover, with this background-free detection technique and time-gated
fluorescence imaging, we have been able to precisely determine the numbers
as well as positions of the transplanted FND-labeled MSCs in organs and
tissues of the miniature pigs after intravenous administration. In general,
the method is applicable to single-cell imaging and quantitative tracking
of other cells in animal models for cell therapy research & development".
